Development of a Dot-Elisa Assay for Diagnosis of Southern Rice Black-Streaked Dwarf Disease in the Field

Outbreaks of the Southern rice black-streaked dwarf virus (SRBSDV) have caused significant losses in many rice-growing areas Vietnam, especially both North and Central Vietnam recent years. To detect virus, traditional reverse transcription polymerase chain reaction (RT-PCR) methodology immunoassays are currently employed. RT-PCR is accurate but requires expensive chemicals instruments, as well complex procedures that limit its applicability for field tests. develop a cheaper, simpler, reliable SRBSDV diagnosis assay based on dot-enzyme-linked immunosorbent (dot-ELISA) method, anti-SRBSDV polyclonal antibodies were produced by using antigens derived from P10 coat protein SRBSDV, which was achieved previous study. The IgG antibody purified antiserum recombinant P10-immunized mice A-agarose affinity chromatography could specifically target at dilution 1:100000. In trial test diagnosis, dot-ELISA obtained showed an accuracy rate 90.9% comparison with standard assay. These results important premises large-scale application order to protect crops against viral disease damage.
